Cellular and Molecular Biology

Infrastructure and Functions
The department comprises several lab-areas in the institute, which provide comprehensive equipment for basic and advanced molecular and cell biology technologies, including working areas under S1 and S2 gene technology safety regulations and for performing experiments with radiolabeled compounds. As in other departments of the institute, lab space and technology platforms are organized into functional units to be shared and used by all current members and visiting scientists at the IKP, in order to maximize financial and organizational efficiency.
Molecular biology working areas (safety level S1)
Standard molecular/biochemical working areas with standard equipment for DNA-, RNA-, and protein-work (standard PCR, gel electrophoresis with digital documentation, Western-Blot station with several units of different sizes, photometer, centrifuges etc.).

Special equipment includes:

  • Beckman-Coulter Table-Top Ultracentrifuges for micro-scale preparations
  • Bioanalyzer (Agilent) for high-resolution DNA, RNA, protein analysis
  • LICOR 4300 DNA Analyzer Laser-Sequencing Station
  • Real-time PCR systems: ABI 7900HT und 7500, Roche 480 LightCycler
  • High-throughput microfluidics PCR platform Fluidigm BioMark HD
  • LICOR Odyssey Fluorescence-based multichannel image-analyzer for Western Blot protein analyis
  • Fuji LAS1000 CCD Camera station for luminescence-Western blotting
  • Enspire (Perkin Elmer) microtiter plate-reader for multifunctional analysis (absorption, fluoroescence, luminescence); includes an AlphaScreen-Module for measurement of biomolecular interactions
  • Autolumat Plus LB 953 (Berthold), automatic chain-based tube luminometer for measuring luminescent reporter gene activities
Cell culture labs (safety levels S1 and S2)
Cell culture work space is organized into separated safety level S1 labs and a safety level S2 lab, which has restricted entry permission. The following functional units and special equipment are provided:

S1 cell culture labs:

  • 5 laminar flow hoods and associated incubators in four S1 cell culture labs
  • Flow Cytometry Analysis Station: Becton Dickinson FACS Calibur
  • InCell Analyzer 1000 High-Content Screening system

S2 cell culture lab:

  • 2 laminar flow hoods (S2-level approved) and associated incubators
  • Autoclave with exhaust-air filter (S2-approved)
  • Ultracentrifuge (Beckman-Coulter) for isoliation of recombinant virus particles
  • Fluorescence cell-culture microscope (Olympus Ck X 41) for observation of GFP/RFP-labeled proteins
Radionuclide laboratory
The radionuclide laboratory provides lab-areas and equipment for working with commonly used radioactive isotopes in cellular and molecular biology. Currently authorized for working with 3H, 14C, 32P, 33P and 35S isotopes, e.g. cellular transport studies using radiolabeled compounds and in vitro DNA-protein and protein-protein interaction studies were performed. Special equipment includes:

  • Liquid scintillation counter Hidex 3000
  • Microtiter plate reader Chameleon for radiometric, fluorometric and luminometric cell based and in vitro assay (e.g. scintillation proximity asays)
  • Hybridization oven
  • BAS 1800 phosphor storage scanner
